BofA Analyst Explains Why Chinese Inflation Actually Fell In March

Headline numbers show that Chinese CPI rose 3.6 percent year-over-year (YoY) in March from 3.2 percent in February. Meanwhile PPI fell 0.3 percent YoY from 0 percent the previous month.

But Ting Lu, China economist for Bank of America-Merrill Lynch, said CPI inflation actually fell in March:

"Before jumping to any conclusions and implications, we should keep the CNY (Chinese New Year) holiday distortion in mind. YoY CPI inflation was upwardly distorted in Jan (at 4.5%) and downward distorted in Feb (at 3.2%) due to the different timing of the CNY holiday. The average (at 3.9%) of Jan and Feb CPI inflation readings is the right measure of inflation in Jan-Feb. In that sense, CPI inflation actually slowed to 3.6% YoY in March from 3.9% in Jan-Feb."

But Lu said CPI inflation could not moderate much further because of increasing price pressures. First, elevated oil prices could impact prices of domestic fuel and chemical products, the government is also likely to seize the opportunity to raise regulated utility prices like water and fuel and finally labor costs are expected to rise.

It is unlikely that China will see a massive stimulus according to Lu, the #1 China economist according to Bloomberg, because he thinks the current growth pace is appropriate for China, and a faster pace would add to inflation pressures.

Lu expects annual CPI to fall to 3.5 percent in 2012, from 5.4 percent in 2011. He expects PPI to fall to 1.7 percent in 2012, from 6 percent in 2011. Both CPI and PPI are expected to fall before rebounding in the second half of the year.

Opening the gate to robust quantum computing: New technique for solid-state quantum info processing

ScienceDaily (Apr. 9, 2012) ? Scientists have overcome a major hurdle facing quantum computing: how to protect quantum information from degradation by the environment while simultaneously performing computation in a solid-state quantum system. The research was reported in the April 5 issue of Nature.

A group led by U.S. Department of Energy's Ames Laboratory physicist Viatsheslav Dobrovitski and including scientists at Delft University of Technology; the University of California, Santa Barbara; and University of Southern California, made this big step forward on the path to using the motions of single electrons and nuclei for quantum information processing. The discovery opens the door to robust quantum computation with solid-state devices and using quantum technologies for magnetic measurements with single-atom precision at nanoscale.

Quantum information processing relies on the combined motion of microscopic elements, such as electrons, nuclei, photons, ions, or tiny oscillating joists. In classical information processing, information is stored and processed in bits, and the data included in each bit is limited to two values (0 or 1), which can be thought of as a light switch being either up or down. But, in a quantum bit, called a qubit, data can be represented by how these qubits orient and move in relationship with each other, introducing the possibility for data expression in many tilts and movements.

This power of quantum information processing also poses a major challenge: even a minor "bump" off course causes qubits to lose data. And qubits tend to interact quite sensitively with their environment, where multiple forces bump them off track.

But, because the key to quantum information processing is in the relationship between qubits, the solution is not as easy as isolating a single qubit from its environment.

"The big step forward here is that we were able to decouple individual qubits from the environment, so they retain their information, while preserving the coupling between the qubits themselves," said Dobrovitski.

Solid-state hybrid systems are useful for quantum information processing because they are made up of different types of qubits that each perform different functions, much like different parts of a car combine to move it down the road. In the case of Dobrovitski's work, the hybrid system includes magnetic moments of an electron and a nucleus.

"This type of hybrid system may be particularly good for quantum information processing because electrons move fast, can be manipulated easily, but they also lose quantum information quickly. Nuclei move very slow, are difficult to manipulate, but they also retain information well," said Dobrovitski. "You can see an analogy between this hybrid quantum system and the parts of a classical computer: the processor works fast but doesn't keep information long, while the memory works slowly but stores information for a long time."

Usually, when you decouple qubits from their environment to protect their quantum data, you decouple them from everything, even from each other.

But, Dobrovitski found a narrow window of opportunity where both the electron and nucleus can be decoupled from their environment, while retaining their relationship to each other.

"The solution is applying a certain pattern of kicks to the electron's magnetic moment, so that tiny rotations between each kick accumulate and coincide with the rotation of the nucleus," said Dobrovitski. "We can separate out this particular single electron movement from thousands of others because it is synchronized with the motion of the nuclear magnetic moment."

As a result, the electron's and nucleus' movements stay linked, while they are both protected from being bumped off course and retain their quantum information processing capabilities.

Experiments carried out by a team of scientists from Delft University of Technology in the Netherlands and University of California, Santa Barbara, showed that theoretical development of this technique worked well in practice.

The researchers took the technique one step further and showed that it can be used for small-scale quantum information processing. Scientists at Delft and UCSB successfully carried out Grover's quantum search algorithm, a method for searching random lists. In this case, they used the solid-state hybrid system to correctly search a list of four random items.

"This is the first time a robust quantum computation has been demonstrated using a solid-state system with individual spins," said Dobrovitski. "We showed that even with the inevitable imperfections of experiments, we can use this system to do quantum information processing in a way that beats its classical counterpart. Indeed, for a list of four items, the quantum device finds with certainty the desired entry by looking into the list only once, while classically we must inspect all four items one by one."

While a four-item list is a small list, consider the possibility of a random list of a million entries. Using classical computing, 500,000 queries would be needed. But, using quantum information processing, only 1,000 queries are required, showing just how much faster tomorrow's quantum information processing will be than today's classical computers.

The research conducted at Ames Laboratory was funded by the DOE's Office of Science.

The Kyocera DuraPlus ($69.99) should come with a heavy machinery warning: If you drop it, it may cause damage to the machinery. The DuraPlus is a gigantic brick of a push-to-talk cell phone, and one of the first to hook into Sprint's next-generation?Direct Connect?service. It's a lot like Kyocera's recent DuraMax?($69.99, 3.5 stars) and DuraCore?($49.99, 3 stars), but trades their flip-style form factor for a slab design that may be easier to use out in the field. The DuraPlus is good for voice-only, push-to-talk communication, but it's a massive phone with few other features.

Design and Call Quality
Colleagues passing my desk kept stopping to comment on the huge size of the DuraPlus. At 5.3 by 2.2 by .9 inches (HWD) and a whopping 6.7 ounces, the DuraPlus is?definitely not going to fit in your pocket, but it'll fit on your tool belt.?Though the virtually indestructible Sonim XP3300 Force?($525, 3.5 stars) is another huge handset, it wears its size more gracefully than the DuraPlus does. The DuraPlus's exterior is made mostly of grippy, scratch-resistant black rubber, with metallic accents on the front.

The DuraPlus is a rugged phone that meets military specification 810G, which makes it resistant to dust, extreme temperatures, low pressure, rain, salt fog, shock, and vibration. It can also be submerged in up to 3 feet of water for 30 minutes. The phone survived numerous drops to the floor of the PCMag Labs, along with half an hour's worth of submersion in a container of water. A plastic screw secures the battery cover tightly, but you can easily unlock it with your fingernail.

The phone's 2-inch, 320-by-240-pixel LCD looks sharp. The font is a little small by default, but you can make it larger. The keypad has too many function keys, but at least all of the keys are large, raised, and well-spaced, which should make it possible to dial while wearing gloves. One of those function keys activates the surprisingly powerful LED flashlight on top of the phone, which may come in handy depending on your work environment.

The DuraPlus is a dual-band EV-DO Rev. A (850/1900 MHz) device with no Wi-Fi.?Reception is average and voice quality is good overall. Voices sound rich and clear in the earpiece, but while the volume goes loud, it isn't loud enough to be heard in a noisy work environment. The speakerphone, however, is very clear and goes super loud, which is something we felt was lacking on the DuraCore and DuraMax. Calls also sounded good through a Jawbone Era?Bluetooth headset ($129, 4.5 stars) and the Nuance-powered voice dialing worked well. Battery life was excellent at 9 hours and 27 minutes of talk time.

Direct Connect
Sprint's new CDMA push-to-talk system combines the fast Internet speeds and nationwide coverage of Sprint's CDMA network with the instantaneous push-to-talk of the old Nextel iDEN network. That's great, but it doesn't really matter for a device like the DuraPlus, which isn't built for heavy Web access.

You need to be in a Sprint coverage area in order to use the DuraPlus, but you can still make push-to-talk calls to Nextel iDEN subscribers.?Direct Connect supports Call Alert With Text, which sends an audio alert and text message to another subscriber, along with Group Connect, which can connect 20 subscribers together at once. You can also use the Direct Connect button to mass-message up to 200 Direct Connect subscribers in one shot, or to send recorded messages to email addresses or handsets via text message.?

For this review, I tested Direct Connect between two different DuraPlus phones. It takes about a second or to initiate the connection, after which transmissions are nearly instantaneous. Voice quality is clear and you should no trouble hearing someone, even in a noisy environment.

Apps, Multimedia, and Conclusions
Rugged form factor and Direct Connect aside, the DuraPlus is an almost featureless feature phone. The main menu is the same 12 icon grid that Sprint has used on many flip phones in the past (including the DuraCore and the DuraMax), which gives the phone a dated look and feel. To make things worse, this interface requires at least nine button presses to perform simple tasks like pairing a Bluetooth headset. There's an Access NetFront 4.1 browser for reading WAP pages, but the screen is too small and browsing is slow. There's support for TeleNav GPS Navigator, which gets you voice-enabled, turn-by-turn directions, as well as Sprint Family Locator.

There's a nonstandard 2.5mm headphone jack on the right side of the phone for voice headsets. The phone has a meager 64MB of free internal memory and no microSD card slot, but without a music or video player, this shouldn't be an issue.

If you need a loud, rugged, brick of a phone to shout commands through, the Kyocera DuraPlus is a decent choice. For the same price, the Kyocera DuraMax is a slightly smaller rugged flip phone, and adds a decent camera and a microSD card slot to the mix. The Kyocera DuraCore costs $20 less, and is essentially the same phone as the DuraMax, minus the camera. If those phones aren't loud enough for you, and you don't mind sticking with Sprint's steadily declining iDEN network, the?Motorola Brute i686?($139.99, 3.5 stars) gets you very loud push-to-talk, albeit for a higher price. And if you're really looking to take advantage of Direct Connect, the Motorola Admiral?($99.99, 4 stars) is a rugged Android smartphone with excellent multimedia features, as well as access to more than 450,000 apps in the Google Play store.

Climbing Mount Immortality: Death, Cognition and the Making of Civilization

Imagine yourself dead. What picture comes to mind? Your funeral with a casket surrounded by family and friends? Complete darkness and void? In either case, you are still conscious and observing the scene. In reality, you can no more envision what it is like to be dead than you can visualize yourself before you were born. Death is cognitively nonexistent, and yet we know it is real because every one of the 100 billion people who lived before us is gone. As Christopher Hitchens told an audience I was in shortly before his death, ?I?m dying, but so are all of you.? Reality check.

In his book Immortality: The Quest to Live Forever and How It Drives Civilization (Crown, 2012), British philosopher and Financial Times essayist Stephen Cave calls this the Mortality Paradox. ?Death therefore presents itself as both inevitable and impossible,? Cave suggests. We see it all around us, and yet ?it involves the end of consciousness, and we cannot consciously simulate what it is like to not be conscious.?

The attempt to resolve the paradox has led to four immortality narratives: Staying alive: ?Like all living systems, we strive to avoid death. The dream of doing so forever?physically, in this world?is the most basic of immortality narratives.? Resurrection: ?The belief that, although we must physically die, nonetheless we can physically rise again with the bodies we knew in life.? Soul: The ?dream of surviving as some kind of spiritual entity.? Legacy: ?More indirect ways of extending ourselves into the future? such as glory, reputation, historical impact or children.

All four fail to deliver everlasting life. Science is nowhere near reengineering the body to stay alive beyond 120 years. Both religi?ous and scientific forms of resurrecting your body succumb to the Transformation Problem (how could you be reassembled just as you were and yet this time be invulnerable to disease and death?) and the Duplication Problem (how would duplicates be different from twins?). ?Even if DigiGod made a perfect copy of you at the end of time,? Case conjectures, ?it would be exactly that: a copy, an entirely new person who just happened to have the same memories and beliefs as you.? The soul hypothesis has been slain by neuroscience showing that the mind (consciousness, mem?ory and personality patterns representing ?you?) cannot exist without the brain. (AP) ? Good Friday was an official holiday in Cuba for the first time in a half century, but few Roman Catholics on the island seemed to be using the day off to attend Mass.

Cuba's Communist government declared the holiday to honor a request that Pope Benedict XVI made during last week's visit. Only a few more worshippers than usual were noted inside Havana's Catholic churches on Friday.

Authorities were also allowing Cardinal Jaime Ortega's to transmit a Good Friday message on state television.

Magno Felipe Mitjans, a lay worker who described himself as "revolutionary, Christian and Catholic," said at his parish of San Juan de Letran, "We have received more people, including in comparison with other Good Fridays."

Good Friday is the day Catholics commemorate the death of Christ, but it is not an official holiday in the United States, most of Europe or even Mexico, the most Catholic of the world's Spanish-speaking countries.

Cuba removed references to atheism from its constitution in the 1990s and relations have since warmed with the church. Still, less than 10 percent of islanders are practicing Catholics. The country has large numbers of adherents to Santeria and evangelical Christianity.

Cubans' low-key commemoration of the day contrasted with observances in other parts of the world.

Roman Catholics and Protestants in the Holy Land commemorated the crucifixion of Jesus Christ with processions through Jerusalem's Old City, and thousands gathered in the Philippines to witness devotees nailed to crosses in a tradition discouraged by the church.

Processions were held around the rest of Latin America, including especially eye-opening ones in Guatemala where streets in some ciites were carpeted with flowers and colored sawdust.

"I'm not Catholic, but I respect them," said Gladys Ocampo, among Cuban workers who got the day off. "I'm happy to have a holiday I wasn't counting on."

Rarest ducklings on Earth hatch

Nigel Jarrett, head of conservation breeding for the WWT, explains why the hatching of the ducklings is such an important conservation milestone

Eighteen Madagascan pochards - the world's most endangered duck - have hatched in a captive breeding centre.

This brings the world population of the ducks to just 60.

The Wildfowl and Wetlands Trust and Durrell Wildlife Conservation Trust, the groups leading the captive breeding programme, say this "builds hope that the bird can be saved from extinction".

The precious pochards are being reared at a specially built centre in Antsohihy, Madagascar.

The ducks were thought to have become extinct in the late 1990s, but were rediscovered in 2006, when conservationists on an expedition spotted just 22 birds at a single site - Lake Matsaborimena (or Red Lake), in northern Madagascar.

Durrell and the WWT launched an emergency mission to rescue the critically endangered birds in 2009.

The aim was to collect eggs in order to start a captive breeding programme that would safeguard the species.

The conservationists collected 24 eggs from nests at the side of the lake. They initially reared the ducklings from those eggs in a hotel bathroom, while a captive breeding centre was being prepared.

The ducks that began their lives in that inauspicious setting have now bred in captivity for the first time.

Dr Glyn Young, a conservation biologist with Durrell, who has spent much of his life studying the Madagascar pochard, said: "The ducklings represent an incredible step forward in the fight to save the pochard from extinction.

"The arrival of these ducklings has led to real hope that the birds can one day flourish again."

'Last hiding place'

Madagascan pochards remain extremely vulnerable to extinction from a single event such as pollution or a disease outbreak.

Scientists are studying the remaining wild population closely, in order to understand the reasons behind the species' decline and to determine the right conditions for releasing birds back into their natural habitat.

Peter Cranswick, head of species recovery at WWT explained that, although Lake Matsaborimena was the "last hiding place for the ducks", it was "far from ideal".

"Our initial investigations suggest there is too little food and this may be leading to the low survival of the ducklings," he said.

"In effect, they are starving to death."

Nigel Jarrett from WWT, who took part in the 2009 rescue mission, explained that the team hoped to find, or even create, wetlands for the birds that were free from the pressures of fishing invasive predators.

He said they hoped to create an environment where the ducks could "survive on their own, and where they can flourish".

Mr Jarrett added that he shared the joy that his colleagues in Madagascar must have felt on seeing the eggs hatch.

"It's the closest thing to becoming a grandparent without being a grandparent," he told BBC Nature.

"When you see a star-shaped crack in an egg and you know there's a living baby inside - a duckling that wants to get out."

The WWT's Idris Bhatti is currently working with the conservation team in Madagascar - helping Malagasy conservationists to learn the skills needed to breed and rear pochards.

He said that "all the effort and hard work checking on the eggs and incubator at least three times in the night and umpteen times in the day" had paid off.

"I felt as if I had laid the eggs and incubated them myself," he said.

"I had incubated and watched eggs hatch hundreds of times before, but on this occasion I couldn't help shed a tear."

The Peregrine Fund, Asity Madagascar and the Madagascan government are also partners in this conservation project.

Michael Martinez has Achieved International Caliber in Ice Skating ...

At a time when most kids his age remain comfy inside the cocoons of parental dependency or otherwise preoccupied with fads or even mischief, Michael has been into rigid training of at least six hours each day ? and school studies in between ? in his efforts to make a better person of himself. In the process he has also been able to put the Philippines in the world ice skating map.

Michael was into recreational skating starting when he was eight-and-a-half years old. He had his first competitive event in Colorado Springs in June 2008 where he garnered several gold medals.

In October 2009 he won gold in all of his first major competitions under the International Skating Union (ISU) in Slovakia, Slovenia and Austria. He is also the undefeated champ of Asian junior figure skating since 2008 and had garnered many other international and local plums.

In September, last year, Michael placed eighth in the Junior Grand Prix of the World Figure Skating Championships held in Brisbane. In June of that same year he received the Hollin?s Trophy in Sydney. In the 1st Winter Youth Olympic Games (WYOG) held from January 16-22, 2012, in Innsbruck, Austria, Michael was third in the short program and seventh in the free skate.

At age 15, Michael had amassed a total of 188 medals and trophies from both local and international competitions ? 146 of them gold medals. He has also a gold medal and five others won in local gymnastics.

Michael was in Belarus early this month competing in the ISU World Junior Figure Skating Championships 2012 and was quite impressed of the venue. In addition to the main ice ring, the huge sports arena has a separate rink for hockey, another for the public to practice, indoor speed skating tracks, medical facilities for athletes and more amenities. There are reportedly two other ice rinks built by the government so that their athletes can train with modern facilities.

?In Philippines,? Michael noted in his Facebook page, ?we only have 1 Olympic size ice rink in the whole country. & our ice rink is inside the shopping mall & it?s mainly for public skating. We are paying ice time but there is no freestyle session, so high level skaters cannot practice there.?

At that time he had just completed the junior men event and was feeling low because of his 15th place in a field of 40 skaters. He should have been ranked higher because his technical scores were the 8th highest, he said. Earlier he placed third in the preliminary rounds and sixth in the short program.

If the above feats and achievements of Michael are not daunting enough, fancy this: Snow in tropical Philippines is as exotic as, say, sighting the mythic Sasquatch. In fact the only ice most Filipinos would see in their lifetimes are the cubed ones we buy to cool our drinks. That being the case, Michael is further handicapped by sub-zero conditions as had occurred in Innsbruck and Belarus. He said of the latter, ?Coming from a tropical country, negative 15 Celsius is too cold for me.?

And then there are the injuries. Michael was quoted in one interview saying, ?Ang ayaw ko lang sa skating: ma-injured.? (The only thing I dislike in skating is getting injured.) And to reduce the incidence of injury, Michael cross-trains in gymnastics, plyometrics and trampoline. Still skating is an injury-prone sport and in addition to being asthmatic, Michael had experienced many falls, repeated injuries and torn ligaments during trainings and competitions.

In Merano, Italy, for example, he had to withdraw from the competition because a thigh was pierced by the skate blade during a bad fall and he had to be rushed to the hospital because of profuse bleeding. In October-November of 2010 he missed several competitions because of ankle injuries that had him in cast for three months. In the recent Belarus games he hurt an ankle which gave him so much pain during practices and programs. Additionally he had sprained a knee and pulled back muscles that required three days of treatment in that country.

Michael is supposed to compete in the Asian Junior Challenge to be held in China this coming April 6 but had to withdraw due to injuries (sprain/swollen ankle) from a very bad fall/twisting of his ankle during his last day of practice in Anaheim before the flight back to Philippines.

Lip service from the government

During a press conference in Nov. 4, 2011, Manuel Veguillas of the Philippine Skating Union (PSU) grandly proclaimed the country?s choice of Michael Martinez as the lone representative to the Olympic Games in Austria: ?We?re blessed to have someone as talented as Michael to represent the country in the Winter Olympics.?

In an exchange of notes with Michael, however, PinoyWatchdog learned that the Philippine government has been nickel-and-diming its support to an athlete who has done much to bring honor and prestige to the country.

Wrote Michael: ?Nung lumaban nga ho ako sa Olympic qualifying last year, I just borrowed the coach of Thailand, kasi wala akong coach hindi kaya ni mama na gumasta para sa coach. Then nung lumaban ho ako sa Youith Olympics last January, wala rin ho akong coach uli. Si mama lang ang tumayong coach ko. Ayaw talaga nilang magsupport. Nung matapos yung Olympics dun lang nagabot ng konting pera ang PSC $700 at shoemart $850 para lang masabing tumulong sila kuno. Its too late dahil dapat tinulungan nila ako sa training ko for Olympics. Then again dito sa world championship, again ni singko wala silang itinulong uli?? (When I competed to qualify for the Olympics last year, I just borrowed the coach of Thailand because my mom could not afford to hire a coach. Then when I competed in the Youth Olympics last January, I again didn?t have a coach. My mother had to stand in as my coach. They really don?t want to give support. It was only after the Olympics that PSC handed out $700 and Shoemart $850 just so it can be said that they were helping. It was too late because they should have helped me during the training for the Olympics. Then again in this world championship, not even five centavos was given to help?)

His mother, Maria Teresa Martinez, agrees. Many times Michael had participated in international competitions without a coach because they could not afford to hire one. In an email dated March 13 to PinoyWatchdog.com, she said: ?I do not know who can help me bring my son?s case to the office of the President. I do not have any more money to support his training, and I feel so bad for my son because he had proven that he can defeat skaters from traditional skating countries, and he wants to qualify & compete in the 2014 regular winter Olympics, but we have no more budget for that. And my other problem was that I was already forewarned by the PSU/NSA that if I bring my complaint of no support to the media, my son may not be given anymore international assignments.?

In another note dated March 22 she said, ?? he (Michael) is not adequately supported/funded by Philippine sports agencies concerned ? the Philippine Skating Union (PSU), the national sports association for figure skating, and the Philippine Sports Commission (PSC). The only money they, PSU & PSC, have given so far was the daily allowance in Innsbruck during the youth Olympic games in Jan., 2012. Phil. Olympic Committee (POC) is helping to get a refund of Michael?s Youth Olympic training expenses from PSC (training expenses in Nov. 7, 2011 till Jan. 7, 2012) but there is no refund until today.?

On May 26, Mrs. Martinez wrote about Michael: ?His skate shoes was so much worn out (he was using it for more than a year, while normally skaters in his level change shoes every 4 to 5 months) but since we were trying to save money on very expensive skate shoes that currently costs $1,200 (with blades) then Michael was forced to skate (and compete in several prestigious competitions including Olympics and world championship) using his worn-out & unstable skate shoes (he was just wrapping the top portion of the shoes with packaging tapes to prevent the shoes from collapsing).?

She is grateful that a donor had come up with the amount to pay for a new pair of skating shoes. The same donor, as well as many other Californians, had also helped in Michael?s U.S. training expenses, she said.

?If Michael was not trained in the U.S., he will never stand a chance in competitive skating,? Mrs. Martinez said. ?But his training abroad depleted our life savings, sad to say. We had also ?exhausted? our relatives and friends who had also been giving financial support in the past, which enabled Michael to compete abroad. Everybody in our family ?sacrifices? just so Michael can continue with his U.S. training. Michael?s sister used to skate too, but had to stop skating due to high cost, same with his 2 nieces.?

?Michael badly needs sponsors for his continuing training in the U.S. since he has more competitions this year starting in July, August and onwards in preparation for his World Championship competition in March, 2013 (which is the qualifying event for the 2014 Winter Olympics),? Mrs. Martinez said. Mother and son Michael are currently in Manila with the latter catching up on school assignments.

Michael is the youngest of three children of Maria Teresa Martinez, a single parent. He will be in fourth year high school of the Muntinlupa School for Child Development in June (home-schooled). The eldest of the brood, Paul Alexis, is 29, married and in charge of the farm business. Maria Angelica, 19, is a full scholar at Colorado College in Colorado Springs pursuing a degree in Economics and Pre-Medicine. A high school graduate from San Beda College in Alabang, Muntinlupa City, she subsequently finished a two-year International Baccalaureate Course at the United World Colleges in Costa Rica.

Death cap mushroom poison to arrest pancreatic cancer in mice

The mere thought of an identification error sends a chill down the spine of any mushroom lover: The death cap mushroom (Amanita phalloides), which resembles the common white button mushroom, contains one of the most deadly poisons found in nature, ?-amanitin. This substance kills any cell without exception, whether it be healthy or cancerous. At the German Cancer Research Center (Deutsches Krebsforschungszentrum, DKFZ) and the National Center for Tumor Diseases Heidelberg, immunologist Dr. Gerhard Moldenhauer, jointly with biochemist Professor Dr. Heinz Faulstich, Max Planck Institute for Medical Research, has now developed a method for destroying cancer cells using the dreaded fungal toxin without harming the body.

The trick to accomplish this is to deliver the poison directly to the right address in the body using something that virtually serves as a cab. In this case, the cab is an antibody whose highly specific arms attach to a cancer-typical cellular surface protein called EpCAM. The fungal toxin is linked to the antibody in a stable chemical conjugation.

In the culture dish, the poison-loaded antibody arrested the growth of pancreatic, colorectal, breast and bile duct cancer cell lines. In mice bearing transplanted human pancreatic cancer, a single antibody injection was sufficient to inhibit tumor growth. Two injections of higher doses of the antibody even caused complete tumor regression in 90 percent of the animals. Even the higher doses did not cause any poison-related damage to the liver or other organs of the animals.

EpCAM, the protein chosen by the Heidelberg immunologists as the tumor cell recognition structure, is a characteristic membrane protein of epithelial cells. This type of cells lines all inner and outer surfaces of the body. Most malignant tumors originate from such epithelial tissues. Many of these, such as pancreatic cancer, breast and ovarian cancers, bile duct carcinomas and tumors of the head and neck, produce too much EpCAM ? and this is frequently associated with an extremely poor prognosis of the disease. EpCAM is therefore considered a suitable target structure for attacking tumor cells.

"Treatments with unconjugated antibodies against EpCAM have already been tested in clinical trials such as for breast cancer. They were intended to attack the cancer solely with the weapons of the immune system, but they turned out to be clinically ineffective," said Gerhard Moldenhauer. "However, our amanitin-conjugated antibody has a much greater potential for killing cancer cells."

Details are of vital importance

Each antibody is linked to between four and eight toxin molecules. Amanitin is regarded as very suitable for this purpose. It is small enough not to be recognized as foreign by immune cells, while it is also robust enough to lend itself to chemical conjugation. "When developing toxin-conjugated antibodies you have to take an awful lot of things into account," Moldenhauer explains. "The cancer cell has to regularly take the target molecule including the attached antibody into its interior, for this is the only place where the poison can act. In the cell's interior, the poison needs to detach from the antibody or else it will not be effective. At the same time it is absolutely vital that it does not get lost while it is being carried through the body, because this could cause severe adverse side effects."

The dosage of the amanitin antibody needs to be determined with the utmost care. One problem is that liver cells are extremely sensitive to the fungal toxin; another is that other healthy cells carry the EpCAM molecule as well and are therefore endangered. However, the results obtained in mice give reason to be optimistic, according to Gerhard Moldenhauer: "Even at high doses we have not detected any organ damage in the animals. We therefore expect that there is a sufficient therapeutic window for a dosage that kills cancer cells while leaving healthy tissue unaffected."

Moldenhauer, who has many years of experience in developing therapeutic antibodies, already has plans for amanitin-conjugated guided missiles against other cancers. In particular, certain types of leukemia and lymphoma cells also carry highly specific surface molecules which lend themselves as target structures for poison-loaded antibodies.

Groupon restatement sparks more worries

(AP) ? Groupon's announcement that its revenue and earnings were lower than what it reported in February is sparking fresh worries about the young company's business model.

Trouble arose when the online deals company said late Friday that it had to revise its previously issued fourth-quarter results to increase the money it set aside for refunds to customers. A big reason: It is selling more expensive deals, such as vacation packages and Botox procedures, which have higher return rates.

Investors responded by selling Groupon's stock on Monday, and shares fell 17 percent. Some analysts are wondering if Groupon, which began trading publicly less than five months ago, will be able to leave behind its growing pains.

"We believe Groupon is now a 'show-me' story, where a company's ambitions are higher than the level of its internal planning and controls," Jordan Rohan, an analyst with Stifel Nicolaus, said in a note to investors. He downgraded the company's stock to "Sell," from "Hold."

"The company claims that the returns do not threaten its expansion and growth," Rohan wrote. "We prefer to wait and see."

Internal controls are systems that companies have in place to make sure that their financial reporting is accurate and that they can meet business goals. Late Friday afternoon ? a popular time for companies to release bad news ? Groupon also disclosed that its auditors found a weakness in its internal controls, but insisted that its business is solid.

Founded in November 2008, Groupon pioneered the online daily deals market, which offers subscribers deep discounts on everything from restaurant meals to tech gadgets to weekend getaways if enough people buy in. It sparked many copycats, including LivingSocial, Google and Facebook, though the latter ended up abandoning the effort.

In terms of mere numbers, the financial restatement doesn't amount to much. The revision lowered the company's fourth-quarter revenue by about 3 percent, to $492 million. And it widened its net loss by 4 cents, to 12 cents per share. Groupon's chief financial officer, Jason Child, said the company remains confident in its business.

But Friday wasn't the first time that Groupon has had to change its accounting. Last year, the company got in trouble for including as revenue the share it had to turn over to merchants running the deal. After federal regulators questioned the fuzzy math, the company counted only the money it got to keep and reduced its reported revenue by roughly half. All that preceded its initial public offering in November.

The fact that Groupon has had to revise its numbers again raises questions about the company's ability to manage its financial reporting and give investors an accurate picture of how it is faring.

Groupon spokesman Paul Taaffe said Monday that the company understands that "there is a certain amount of skepticism in the market" given the company's history.

But he said the company's fundamental business is strong, and such accounting issues are just part of being a business that is growing very quickly. Groupon's revenue nearly tripled in the fourth quarter from a year earlier, and its active subscriber base grew fourfold to 33 million.

"Every three months, Groupon is a different company," he said.

Groupon has evolved quickly from its roots offering mostly restaurant and spa deals from local merchants. It now curates vacation packages to such places as Egypt and the Horseshoe Casino in Robinsonville, Miss., and it sells consumer products such as memory foam mattresses and noise-canceling headphones.

Its deals aren't really daily any more, either. Subscribers can sign up for several email lists from Groupon, receiving multiple deals a day. They often have several days to decide whether they want to buy them.

More recently, with products such as an online scheduling tool for local merchants, Groupon has also shown that it wants to be an e-commerce hub for small businesses.

Several analysts are giving Groupon the benefit of the doubt even as they note shareholder frustrations. Dough Anmuth of JPMorgan said he doesn't believe the restatement shows that Groupon's business model is weakening.

"Instead we believe they reflect Groupon's dynamic deal mix and the company's extremely rapid growth in a very short amount of time," he said.

Scott Devitt of Morgan Stanley, the main underwriter for Groupon's IPO, called the announcement a "mild hiccup in Groupon's compelling long-term story."

The Chicago-based company's stock fell $3.11, or 16.9 percent, to close Monday at $15.28. The stock price is 24 percent below its IPO pricing of $20 and has traded from $14.85 to $31.14.
